There is a contrast between "treatment reduces postoperative pain" and "one started after surgery." because of the "than".
one started after surgery means - after surgery
so "treatment reduces postoperative pain" means before/during surgery.
so first blank goes with preemptive.
Second blank says failed to demonstrate - I do not have a proper clue for this . But by POE it should be something positive . I actually guessed "effectively".
so one that closely matches it would be unequivocably.
