The meaning of the sentence is the following

he was a comedian so the directors were reluctant to employ him for a role at the opposite of his nature as actor.

So C is the only answer that fits the ballpark.

Notice how the sentence implies in both cases a lead role. So, for instance, A is wrong

'Loath' means to be reluctant. So, because of his success as a comedian, the director would not want him into dramatic roles.
